ABSTRACT

A lung diffusion function detection system is designed. Firstly, the controllable collection of air, test gas source and calibration gas source was based on single-breath method measurement principle. Secondly, pulmonary diffusing capacity for carbon monoxide (DlCO) was calculated by gas concentration measured by the non-dispersive infrared sensor to measure, the gas flow measured by the differential pressure sensor, and the temperature, humidity and atmospheric pressure sensors to test and evaluate the quantitative detection and evaluation of lung diffusion function. Moreover, a preliminary verification of the lung diffusion function detection system was implemented, and the results showed that the error of the lung carbon monoxide diffusion and the alveolar volume did not exceed 5%. Therefore, the system has high accuracy and is of great value for early screening and accurate assessment of COPD.

ABSTRACT

Indirect energy metabolism measurement is the gold standard for providing nutritional support for critical illness. The accuracy of the measurement data directly affects the outcome of the disease. In order to study the influence of sampling delay on the accuracy of energy metabolism measurement under mechanical ventilation, the Matlab/Simulink platform and respiratory electrical model were used for simulation and quantitative analysis. The results show that the error of indirect energy metabolism measurement increases with the increase of sampling delay, the error of sampling delay in mechanical ventilation mode is larger than that of spontaneous breathing, and the error of sampling delay in PCV mode of mechanical ventilation is larger than that in VCV mode. Therefore, there should be different sampling delay compensation strategies under severe mechanical ventilation and its different control modes.

ABSTRACT

In order to detect freezing of gait of Parkinson's patients automatically, a system based on inertial measurement unit to detect freezing of gait for Parkinson's patients is established. The two inertial measurement units are respectively fixed on the left and right ankles of the patient to be measured, the freezing index is calculated by windowed Fourier transform, the freezing threshold is calculated based on the freezing index during normal walking, and the freezing index and the freezing threshold are compared to complete the detection of freezing of gait. The experimental results show that the number of freezing of gait occurrences in Parkinson's patients is accurately detected, and it has high sensitivity and specificity, which can assist doctors to objectively assess the patient's condition.

ABSTRACT

OBJECTIVE@#This paper used plantar pressure analysis equipment to discuss the plantar pressure distribution pattern and balance ability of patients with the knee joint injury under static standing.@*METHODS@#Zebris FDM-S plantar pressure analysis equipment was used to collect plantar pressure data from subjects with the knee joint injury and healthy control subjects. We compared the pressure values in each region of pelma, and then assessing the balance ability of the subjects based on the trajectory of the overall pressure center movement.@*RESULTS@#Compared with the healthy control group, patients with knee joint injury have a significantly lower pressure in the heel areas and a significantly higher pressure in the middle foot area. And the total pressure of the injured limb foot in the patient group is lower than that in the contralateral foot. In addition, there is a significant increase in the parameters of the balance ability index in the patient group.@*CONCLUSIONS@#The plantar pressure distribution pattern of patients with knee joint injury is different from that of normal people in static standing, and the balance ability is poor. Therefore, this method has a positive guiding significance in the evaluation and application of knee joint injury.

ABSTRACT

Objective To investigate the variation characteristics of adenovirus type 55 (HAdV-B55) gene on plateaus.Methods Throat swabs were collected from HAdV-B55 infected patients and used for virus isolation in HEp-2 cells.The whole-genome sequence was obtained by PCR and sequencing.HAdV-B55 gene sequence was blast with the previously reported virus.Results HAdV-B55 strains were isolated from throat swabs, which were named LS89/Tibet/2016.The whole-genome sequence was obtained and submitted to GenBank with the accession number of KY002683.No large fragment gene recombination was found between this HAdV-B55 strain and previous strains, and the sequence similarity with QS-DLL strain was 99.9%.Conclusion This study provides more information for the evolution patterns of adenovirus 55 and will contribute to the prevention and control of HAdV-B55 infection in the future.

BACKGROUND: Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) is not applicable for clinical fast detection or the detection analysis of multiple genes on a large sample basis for candidate genes of mental diseases, while polymerase chain reaction-sequence specific primer (PCR-SSP) method can make up for these deficiencies.OBJECTIVE: To establish a PCR-SSP method with multiple gene polymorphism, and popularize its application in the molecular study of mental diseases.DESIGN: Observational trial.SETTING: Department of Molecular Biology and Department of Pathophysiology at Xinxiang Medical College.MATERIALS: The experiment was carried out in the Department of Molecular Biology at Xinxiang Medical College from December 2005 to October 2006. A total of 200 blood samples from patients with mental disease were provided by the Second Affiliated Hospital of Xinxiang Medical College, 0.5 mL each sample. All the patients were informed and agreed with the experiment.METHODS: PCR-SSP was used to analyze the polymorphism of the following genes: variation of tumor necrosis factor (TNF)-α-308A/G and -238G/A, variation of interleukin (IL)-6-174G/C, variation of the 188th base pair (C/T) of CYP2D6 *10B exon, variation of the 472nd base pair (G/A) of COMT gene, and variation of the 941st base pair (G/T) of the CDNA sequence of the 8th exon of MAOA gene.MAIN OUTCOME MEASURES: Gene polymorphism of blood samples analyzed with PCR-SSP.RESULTS: The detected genotypes for TNF-α-308NG included G/G and A/G; the detected genotypes for TNF-α-238G/A included G/A, G/G and A/A; the detected genotypes for IL-6-174G/C were all G/G homozygotes; the detected genotypes for the 188th base pair (C/T) of CYP2D6*10B exon included C/C, C/T and T/T; the detected genotypes for the 472nd base pair (G/A) of COMT included A/G, A/A and G/G; the detected genotypes for the 941st base pair (G/T) of CDNA sequence of the 8th exon of MAOA included T/G, T/T and G/G.CONCLUSION: PCR-SSP is suitable for the polymorphism analysis of the mononucleotide site variation of multiple genes on a large sample basis, which is fast and accurate with low cost and is worthy to be popularized in the molecular study on mental diseases.

ABSTRACT

Objective To study the effect of human ?-globin matrix attachment region(MAR) on transgene expression in stably transfected CHO cells.Methods Expression vector was constructed,which contained the ?-globin MAR in both sides of Chloramphenicol acetyltransferase(CAT) reporter gene expression cassette in cis,then transfected into CHO cells.The CAT reporter gene expression was analyzed by ELISA method.Results The ?-globin MAR enhanced the CAT gene expression 5.5-fold in stably transformed CHO cells,while the transgene expression variation among individuals of transformants was decreased.Conclusion MAR increase transgene expression in stably transfected CHO cells.

Objective To investigate the change in potassium channel in ventricular myocytes in vitro derived from diabetic rats and the effect of insulin and dichloroaectic acid(DCA). Methods The diabetic rat model was established by injection of streptozocin(STZ) peritoneally using male Sprague-Dawley rats with body weight 150-200 gram.
